OpenCRISPR-1 is a fully AI-generated genome editor from Berkeley startup Profluent Bio, consisting of a Cas9-like protein and matching guide RNA, entirely designed using Profluent's large language models. The protein sequence is hundreds of mutations away from natural SpCas9 yet achieves comparable editing activity with roughly 95% fewer off-target cuts and lower immunogenicity. The system was released as a preprint and open-source release in April 2024 and peer-reviewed published in Nature in July 2025; sequences are freely available via GitHub and Addgene.
